前天从美国在线，BBC在线发现一个消息，说视网膜色素病变有了基因疗法，美国已经有三例20岁左右的年轻人接受了治疗，效果很理想，夜间视力也大大提高。英国去年也作了类似治疗试验，也报告了好效果。请问是不是真的？大概多久可以应用临床？谢谢。

临床正式批准可能仍然需要时间（05，04年好像在老鼠身上），因为下一步是将剂量加大植入...之后要大规模临床多期实验，在确定没有危险的情况下才行。英国作了三个人，前面两个没有最后这个效果好，不过起码阻止/缓慢了病情，最后这个比较振奋人心。

The operation involves injecting fluid with missing gene within a modified virus into the eye.

A fine needle (cannula) is passed through the front of the eye and across the vitreous gel.

The cannula is pushed through the retina, light sensitive tissue that lines the back of the eye.

The fluid is injected beneath the retina, causing it to detach from the underlying pigment layer.

Cells in the pigment layer absorb the fluid and the retina returns to its normal position.

The virus infects cells of pigment layer, supplying the gene required for normal sight.

基本原理是利用重组腺相关病毒(Adeno Associated Virus)介导，向患者的眼睛提供健康的RPE65基因，但病毒本身对于人們不会引起任何症狀（因为人体中本来就有很多腺相关病毒的存在）。

[B]Gene therapy now effective in retinitis pigmentosa[/B]

Now there is a good news for thousands of patients who are suffering from Leber congenital amaurosis type 2, as recent news confirms the success of gene therapy on three patients.

The news relates the regaining of the eye sight of three people, one women and two men from 21 to 24 years old who were suffering from a rare, incurable form of hereditary blindness called Leber congenital amaurosis type 2 (LCA2) through gene therapy.

LCA2 (retinitis pigmentosa) is a disease in which the photoreceptor cells of human body fails to respond to light. This is due to a gene called RPE65 which fails to produce a protein that have an importance in healthy vision.

In the study, researchers used an adeno-associated virus, a harmless virus which already exists in most of the people, to deliver RPE65 to a small area of the retina which ultimately helped in the formation of the protein that helps in healthy vision.

This study is a milestone, as this new treatment will be effective in treating human vision disease which are inherited in families and results from mutations in single genes, says Barry J. Byrne, M.D., Ph.D., a professor of molecular genetics and microbiology and director of UF's Powell Gene Therapy Center,

【大紀元9月25日訊】〔自由時報編譯羅彥傑／綜合報導〕英國每日郵報二十四日報導說，直接將健康基因注入病患眼睛的視網膜內，可能讓罹患李柏氏先天性黑矇症的盲人重見光明，而且對其他遺傳性眼疾患者也很有效，尤其是能進一步提升老年性黃斑部病變的治療效果。

李柏氏先天性黑矇症患者福音

美國費城賓州大學施艾氏眼科研究所的科學家，替三名罹患李柏氏先天性黑矇症的二十多歲病患進行試驗性治療。這種先天性疾病會讓患者在十多歲的年紀失明，發病原因在於RPE65基因有缺陷，造成眼球後面的視網膜細胞停止運作。此症在此試驗前一直無藥可醫，但在科學家施以新療法後的短短一個月內，三人的視力獲得大幅改善。

數月前，英國倫敦莫菲爾眼科醫院的外科醫師也是用相同療程，將數百萬個健康基因注射到其中一個視網膜內，幫助一名少年恢復部分視力。這名少年說：「一開始我眼睛完全看不見，但後來漸入佳境，最後恢復正常。」

受到此一突破的鼓舞，賓大科學家再找三名病患來測試此一療法。在短短數天內，其中兩人感覺到視力有改善；一個月後，三人都能看得更清楚。相關研究成果刊登在最新一期的「美國國家科學院院刊」。

科學家發現，患者視網膜的感光視錐細胞與視桿細胞有大幅改善。負責色彩與白天視力的視錐細胞，功能增加達五十倍；提供夜間視力的視桿細胞，效果則大增逾六萬三千倍。但科學家也注意到，患者要比平常花更多的時間才能在黑暗中看見東西，而且要以好幾個小時才能讓眼睛適應環境。

他們指出：「這些結果顯示，在進行RPE65基因療法後，儘管仍有缺憾，但視桿細胞與視錐細胞的光感受體會急遽恢復。」

科學家相信，這項新療法在兩年內就可能用來治療視網膜的其他先天性疾病，五年之內就能嘗試用在老年性黃斑部病變患者身上。(http://www.dajiyuan.com)

411老师,犹如配型的干细胞能彻底解决白血病一样, 有无可能利用干细胞再造硬化的肝脏呢? 国外有无类似研究和进展呢?
