AbstractThe elimination of viral covalently closed circular DNA (cccDNA) from the nucleus of infected hepatocytes is an obstacle to achieving sustained viral clearance during antiviral therapy of chronic hepatitis B virus (HBV) infection. The aim of our study was to determine whether treatment with siRNA is able to suppress viral cccDNA amplification using a HBV-transgenic mice model. The experimental results revealed that siRNAs can serve as efficient alternative anti-HBV agents, because they showed better inhibitory effect on viral replication and antigen expression in transgenic mice. More importantly, the siRNA markedly inhibited HBV cccDNA amplification.

摘要
病毒共价闭合环状DNA的消除(cccDNA的)从被感染的肝细胞的细胞核是一个障碍中的慢性乙型肝炎病毒(HBV)感染的抗病毒治疗实现持续的病毒清除。本研究的目的是确定用siRNA治疗是否能够使用的HBV转基因小鼠模型中抑制病毒的cccDNA扩增。实验结果表明,siRNA的可以作为有效的替代抗HBV剂,因为它们表现出对病毒复制和抗原表达的转基因小鼠更好的抑制效果。更重要的是,所述siRNA明显地抑制HBV的cccDNA扩增。
