"...Two experimental hepatitis B virus (HBV) drugs and assay methods were described in some detail. Sacks et al. (abst. 12) discussed the use of a real time PCR method to assess the level of covalently closed circular (ccc) HBV DNA in the woodchuck model. CccDNA is a virus transcriptional template important to maintenance of chronic infection. The detection system was used to evaluate Clevudine (L-FMAU) therapy in the woodchuck infection model. Korba et al. (abst 13) described a strategy used in the woodchuck HBV system to decrease virus load by Clevudine treatment, followed by virus s-antigen immune stimulation to delay disease development. Several papers also described development of the L-nucleosides (LdT and LdC) for the treatment of HBV infections (Juodawlkis et al. abst. 14; Cretton-Scott, et al. abst. 16; Benzaria et al. abst. 137; Pierra et al. abst. 138; Bridges et al. abst. 146). LdT and LdC together have shown significant synergy, both in vitro and in the woodchuck HBV infection model. The drugs are non-mutagenic and demonstrate no apparent effects on mitochondrial DNA synthesis. In vivo efficacy is observed using 10 mg/kg/day. The oral bioavailability of LdC is relatively poor in woodchucks (~10%) and monkeys (~16%), yet the high drug potency still results in efficacy in the woodchuck model. By developing valyl prodrugs of LdC, the bioavailability is at least doubled and the plasma concentration is increased about 3-fold.
